To the Editor: Melanoma on chronically sun-damaged skin (MSDS) is challenging to detect.1Sanchez M.I. Rabinovitz H.S. Oliverio M.C. Elgart G.W. Grichnik J.M. Our objective was determine whether existing technology could be used create 3-dimensional (3D) models, which would expand data available for discriminating biology. Melanocytic cells have different morphologic appearances dependent developmental stage, activation status, and pathogenic genetic epigenetic alterations. Cell shape, therefore, can provide clues a cell's behavior. We reconstructed individual melanoma from stacks RCM images (Fig 1; Supplemental Fig 1, via Mendeley at https://data.mendeley.com/datasets/7kx38d9h2v/3). Distinct morphologies identified, including spindle, spheroid, heavily dendritic cell forms. These 3D reconstructions also allowed modeling nuclear shapes insets). Because image sizes are known, it possible calculate cellular volumes, may increase accuracy.2Chang C.K. Tsai C.C. Confocal microscopy is used in the study of colloidal gels, glasses, and binary fluids. We measure the three-dimensional positions of colloidal particles with a precision of approximately 50 nm (a small fraction of each particle's radius) and with a time resolution sufficient for tracking the thermal motions of several thousand particles at once. This information allows us to characterize the s...
متن کاملThree Dimensional Statistical Image Analysis and Confocal Microscopy
An important area of interest in microscopy may be the shape and spatial arrangement of the objects under study, aspects such size, interaction and orientation. Confocal fluorescence microscopy is a useful device for imaging three dimensional specimens because of its non-destructive and non-invasive nature which preserves such spatial information. However, imaging problems still arise; low sign...
متن کاملConfocal Brillouin microscopy for three-dimensional mechanical imaging.
Acoustically induced inelastic light scattering, first reported in 1922 by Brillouin1, allows non-contact, direct readout of the viscoelastic properties of a material and has widely been investigated for material characterization2, structural monitoring3 and environmental sensing4. Extending the Brillouin technique from point sampling spectroscopy to imaging modality5 would open up new possibil...
متن کاملThree-dimensional image sensing by chromatic confocal microscopy.
In the image of a confocal microscope, only those parts of an object appear bright that are located in the focal plane of the objective. Because of an axial chromatic aberration deliberately introduced into the microscope objective, the location of the focal plane depends on the wavelength used. By using a white-light source and examining an object with a depth variation less than the axial ran...
